Pudji Astuti, MP</creator><subject>ETD</subject><description>Handling of slaughter cows in abattoir which violates the animal welfare can cause stress and decrease the meat quality. Nowadays the measure of stress hormone is still expensive and less practical, thus it uses another method namely Fourier Transform Infrared Spectroscopy (FTIR). This research is aimed to find out the comparison process of slaughter between rope casting local (RCL) and restraining box mark 1 (RBM 1) observed by the stress level using FTIR method. This research employs 30 male beef cattle Ongole breeds which are divided into two groups, 15 male beef cattle using rope casting local in abattoir CV. Restu Bumi, Segoroyoso, Pleret, Sewon, Bantul, Yogyakarta and 15 others using restraining box mark 1 in abattoir Giwangan, Bantul, Yogyakarta. Blood sampling is taken in jugular vein when they rest, whereas when they are slaughtered, it is taken by collecting the blood out. Velocity of centrifuged blood is in 3000 rpm or 1500 g for 15 minutes to get the serum, then it is measured at wavelength between 4000-400 cm&#xE0;&#xAC;&#xBF;&#xE0;&#xAC;&#xB5; using MB3000 FTIR. Band data absorption in wavelength of FTIR is analyzed descriptively by using FTIR HorizonMB&#xE0;&#xAD;&#xFFFD;&#xE0;&#xAD;&#xFFFD; . Confirmation test of hormone level employs Enzyme Linked Immunnosorbent Assay (ELISA). Statistic test of Mann-Whitney Test is to find out the different mean of cortisol level at two process of slaughter. Result of FTIR research with the wavelength has band absorption of methyl group (= CH&#xE0;&#xAC;&#xB7;) 2986 cm&#xE0;&#xAC;&#xBF;&#xE0;&#xAC;&#xB5;, methylene (= CH&#xE0;&#xAC;&#xB6;) 2827 cm&#xE0;&#xAC;&#xBF;&#xE0;&#xAC;&#xB5;, hydroxyl (-OH) 3371cm&#xE0;&#xAC;&#xBF;&#xE0;&#xAC;&#xB5;, carbonyl (ketones) (C=O) 1636 cm&#xE0;&#xAC;&#xBF;&#xE0;&#xAC;&#xB5;, carboxyl (COO&#xE0;&#xAC;&#xBF;) 1408 cm&#xE0;&#xAC;&#xBF;&#xE0;&#xAC;&#xB5;, glucosa 1057 cm&#xE0;&#xAC;&#xBF;&#xE0;&#xAC;&#xB5;, urea 1011 cm&#xE0;&#xAC;&#xBF;&#xE0;&#xAC;&#xB5;. Cortisol level mean measured with ELISA technique of slaughter employing RCL (23,27 &#xC2;&#xB1; 15,68 ng/mL) is significantly different (P&lt;0.05) with RBM 1 (53,47 &#xC2;&#xB1; 40,45 ng/mL), thus it can be concluded that stress level of RCL slaughter process is lower than RBM 1. FTIR is really possible to be used as stub stress detector that can differ the slaughter process between RCL and RBM 1 by recognizing the increase of absorption and the separation of component group in cortisol and serum at the wavelength because of stress influence.</description><publisher>[Yogyakarta] : Universitas Gadjah Mada</publisher><date>2014</date><type>Thesis:Thesis</type><type>PeerReview:NonPeerReviewed</type><identifier> , C.
