- Escherichia coli becomes the cause of diarrhea because it is carried by the vector animals namely flies, as insects that included in the Diptera order. Flies are annoying insects because their preferences perch in damp and dirty places such as landfills, markets, final waste collection sites (TPA), temporary dumps (TPS) and residential areas. The purpose of this study was to analyze the amount of Escherichia coli found in various types of flies in the residential area and TPST Mulyoagung, also to analyze the use of the research results as a practical guidebook. The study was divided into 2 stages, the stage I of the activity was the comparison of Escherichia coli in various types of flies in the residential area and TPST Mulyoagung Malang, and the stage II was the study of the development of research results into a practical guidebook. The type of research activity in stage I was facto exposures with an observational approach. For the stage II was focus on the development studies using the 3E Learning Cycle model. The 3E Learning Cycle model consists of three stages, namely, the exploration stage (exploration), the concept explanation stage (explanation) and the elaboration stage (elaboration). Flies that taken from the residential area and TPST Mulyoagung were used as samples, obtained 3 types of flies namely Musca domestica, Chrysomya megacephala and Droshopilla sp, then the isolation process was carried out with dilution of flies to10-3and grown in a manner of strake plates on EMB Agar selective media. Identification of Escherichia coli was carried out by direct vision where Escherichia coli colonies were golden green like metal flashes (metallic sheen) and the calculation of the number of Escherichia coli using colony counter with units (CFU/ml). The data analysis technique that used in this study was Two-way Variant Analysis and Duncan Test 5%. The results showed that the highest number of Escherichia coli in Musca domestica in TPST Mulyoagung was 217x103 CFU/ml and the lowest number in Droshopilla sp in residential area was 13x102 CFU/ml. The results of this study were developed in the form of a practical guidebook for Class X Senior High School.
