  • Apple (Malus sylvestris Mill) is one of the fresh fruit consumed by many people. Department of Agriculture (2010) stated that in 2000 the number of apple trees in Batu city was 3,107,195 trees with total production of 147,000 tons per year. However, in 2004 decreased by 1 million trees that decreased local apple production as much as 46 thousand tons per year. One way to increase local apple production is by indirect in vitro culture technique, that is through the stages of embryo formation in apple callus whose process is called embryogenesis. Somatic embryogenesis is the process of shoot formation without going through gamete fusion. Giving sugar one of the organic components into the growing medium serves as a carbon source, maintaining the osmotic pressure of the medium, since most parts of the plant or eksplan that are cultured are not autotrophic and have a low rate of photosynthesis. This study aims to examine the right sugar combinations that affect somatic embryo induction. This research was conducted by using Factorial Randomized Complete Design (RAL) with two factors of treatment that is the combination of sugar (sucrose, glucose, fructose) and apple varieties treated repeated three times. The data can be analyzed variously and in real honest appeal test (BNJ) 5% level. The results showed that the color change in combination of 20gr / l sucrose + 10gr / l glucose with Rome Beauty varieties gave a significant effect on the color parameter of callus 30 HST of 6.67% aged 60 HST of 5.78%. fresh callus of 0.52 g, weight increase of 0.44 g of callus to the induction of somatic embryo apple kalus.
