Kirby-Bauer Test Ekstrak Peras Buah Maja Matang sebagai Inhibitor Urease dan Inhibitor Nitrifikasi pada E.coli

  • Ammonia (NH3) and nitrous oxide (N2O) gas emissions are the two gases produced by the decomposition of urea such animal urine fertilizer. The change urea to ammonia by environmental temperature and microorganism activity is the process of losing soil nitrogen (N) sources and causing unpleasant odors. Further NH3 change to N2O, one of the greenhouse gas (GHG) triggered by nitrification activities of microorganisms that is one of the modifiers of atmosphere balance. This study aims to prevent of nitrogen loss to ammonia on urine: cattle, dairy cows, goats, sheep, and N2O from nitrification of sodium nitrite (NaNO2) and ammonium sulfate (NH4)2SO4) by using mature Maja fruit juice extract (EPBMM) with absorbent paper and liquid as urease inhibitor (IU) and nitrification inhibitor (IN) Escherichia coli grown on 1/10 nutrient medium in solid and liquid forms. 100% EPBMM concentration positively inhibits E.coli growth at 5% concentration of all urine, 0.02% NaNO2, and (NH4)2SO4. EPBMM as IU and IN decreases in amount of the concentration used 20%, 10%, and 5% indicating by Kirby-Bauer Test. Proceeds by the same test, 100% EPBMM inhibition does not decrease after being heated to 117oC. EPBMM properties is blackish brown, smells fresh, and pH 4. EPBMM acid properties can decrease the pH value to prevent urea turning into NH3 and ammonium to N2O. The results of this test became the based knowledge of EPBMM as IU and IN on E.coli which is a useful natural product to protect soil from losing nitrogen to NH3 and N2O.
