  • Mahkota dewa is one Indonesian medicinal plants which is used for treatment of various diseases. The aim of this research is to isolate secondary metabolites from antioxidant active extract of fruit, bark and leave of mahkota dewa and to determine the molecular structure of isolated compound using spectroscopic method and to know the antioxidant activity of the isolated compound containing phenol group. Isolation of secondary metabolites from fruit, bark and leave of mahkota dewa were initiated by extraction using methanol as solvent. After concentrated, crude methanol extract was extracted using n-hexane, chloroform and ethyl acetate, respectively. All of extracts, except n-hexane extract were analyzed their antioxidant activity by DPPH method. Chloroform, ethyl acetate and methanol extracts from fruit, bark and leave that have antioxidant activity were purified using column chromatography. The fraction of column chromatography were purified further by recrystalization and or recolumn chromatography and or TLC preparative. Structure elucidation was performed by spectroscopy using UV, IR, 1H NMR,13C NMR, HMBC, HMQC, COSY, DEPT and MS. Isolated compound which contained phenolic group were analyzed their antioxidant activity by DPPH method. Spectroscopy analysis of phenolic compound with many C chiral atoms was supported by prediction using a series of computational calculation with Hyperchem using semi empirical method. This work resulted 7 isolated compounds i.e. glucoside benzophenone from ethyl acetate extract of fruit and 6 new compounds. New compounds are naphtoic acid derivative and isophalerin A from ethyl acetate extract of fruit, glycoside diterpenoid (phaleriaelide) from methanol extract of fruit, diepoxylignan (macronone) from ethyl acetate extract of bark, aglycon benzophenone and isophalerin B from ethyl acetate extract of leave. Antioxidant activity of the isolated of phenolic compound was benzophenone aglycon (very strong, IC50 10,57 Î1⁄4g/mL), macronone (weak, IC50 240,14 Î1⁄4g/mL), isophalerin B (weak, IC50 402,70 Î1⁄4g/mL) and benzophenone glucoside (no have antioxidant activity). Based on Hyperchem with semiempirical PM3 method, stereoisomer of macronone is lignan 14 which gave similar result with 13C-NMR data.
