- Mangosteen pericarp (Garcinia mangostana L.) and betel leaf (Piper betle L.) has antibacterial activity against Streptococcus mutans due to xanthones and phenol content. This research aims to know the concentration of PVP as a binder and mannitol as a filler a which can produce lozenges formulation optimization. Lozenges were made in 4 formula with combination PVP 3% and 5%-mannitol 70% and 80%. Antibacterial test lozenges using agar cup diffusion methods. The methods formulation optimization used is factorial design and verified using test-t with 95% confidence level. Optimum formula from the proportion of PVP 125 mg and manitol 1982,55 mg able to decrease flow rate granule, decrease hardness, decrease friability tablet, increase time dissolves, taste response, and inhibition zone diameter lozenges. Verifiation results one way t-test level of 95% showed significant difference to outcome prediction for friability and time dissolves. The result optimum lozenges obtained brownish yellow, typical odor betel, flow time of 9,8 seconds, hardness of 12,22 kg, friability of 0,31%, dissolves time of 12,05 minutes, taste response of 5,75, and inhibition zone diameter against the bacteria Streptococcus mutan of 13,5 mm.
