  • Klebsiella pneumoniae adalah bakteri batang gram negatif yang merupakan patogen sangat infeksius. Mekanisme virulensi K. pneumoniae masih belum sepenuhnya diketahui terutama kemampuannya sebagai protein hemaglutinin dan protein adhesin. Tujuan penelitian ini adalah mengetahui peran outer membrane protein (OMP) 20 kDa bakteri K. pneumoniae sebagai protien hemaglutinin dan adhesin. Penelitian ekperimental ini dilakukan pada bulan Oktober – Desember 2018 di Laboratorium Mikrobiologi Fakultas Kedokteran dan Fakultas MIPA Universitas Jember. Dilakukan identifikasi berat molekul OMP dengan 12,5% SDS-PAGE. OMP dengan berat molekul 20 kDa yang diperoleh kemudian digunakan untuk uji hemaglutinasi dan uji adhesi. Hasil penelitian didapatkan OMP 20 kDa K. pneumoniae merupakan protein hemaglutinin dan protein adhesin. Analisis dengan korelasi Pearson menunjukkan terdapat hubungan antara dosis OMP 20 kDa K. pneumoniae dengan indeks adhesi (p= 0,029; R= -0.562). Kata kunci: Klebsiella pneumoniae, OMP, hemaglutinin, adhesin.   Virulence factor of 20 kDa Outer Membrane Protein (OMP) Klebsiella pneumoniae as Haemagglutinin and Adhesin Protein   Abstract Klebsiella pneumoniae is a Gram-negative rod bacterium, which is an exceptionally infectious pathogen. The mechanism of virulence of K. pneumoniae is still not fully known, especially its ability as a hemagglutinin protein and adhesin protein. The purpose of this study was to determine the role of the outer membrane protein (OMP) of 20 kDa K. pneumoniae bacteria as hemagglutinin and adhesin protein. This experimental research conducted in October - December 2018 in the Microbiology Laboratory of the Faculty of Medicine and the Faculty of Mathematics and Natural Sciences, University of Jember. Outer membrane protein molecular weight identified with 12.5% SDS-PAGE. The OMP with a molecular weight of 20 kDa obtained then used for hemagglutination and adhesion tests. The results showed that 20 kDa K. pneumoniae OMP is a hemagglutinin protein and adhesin protein. Analysis with Pearson correlation shows there is a relationship between the OMP dose of 20 kDa K. pneumoniae and adhesion index (p = 0.029; R = -0.562).
