  • Nanoparticle preparation of papaya leaves’ ethanolic extract (Carica papaya L.) and lactic acid isolate from kumpai legum aimed to increase the extract’sactivity as antidiarrhea so it can inhibit diarrhea disease caused by Escherichia coli bacteria. Preparation of papaya leaves extract and lactic acid isolates from kumpai legumes into nanoparticles was using ionic gelation method. Three formulas (F) in this study were evaluatedon the variations ofconcentrations of Ca(OH)2isolates, 12.5 mg for F1, 17.5 mg for F2, and 22.5 mg for F3. The three formulas were resulting encapsulation efficiency (% EE) respectively of 74,58 ± 0,005; 74,51 ± 0,011; dan 74,41 ± 0,015%. Formula 1 resulted the highest EE% so it was selected as the optimum formula. The result of nanoparticles characterizationsuch as average diameter and PDI (Poly Dispersity Index) using PSA (Particle Size Analyzer) in optimum formula were 1444,8 nm and 0,199 respectively. The results of antidiarrheal activity test on optimum formula with dose of 1 gram extract showed that nanoparticle preparation had antidiarrheal activity equal to quercetin 10 mg/kgBW against diarrhea induced byE.coli(p> 0,05) and more effective for weight improvement, fecal consistency and diameter, as well as the time range of diarrhea in mice.
