- Background: Cardiovascular disease is the number one cause of death in the world. The death rate caused by cardiovascular disease in 2015 reached 17.7 million, representing 31% of all deaths in the world. Gotu kola is a medicinal plant that is efficacious because of the active ingredients contained in it, especially triterpen saponisida such as asiatika acid, madecassic acid, asiaticosida, and madecassosida. In previous studies it has been proved that Centella asiatica (L.) has a protective effect on neuron cells through the modulation of acetylcholine. This study aims to determine the efficacy of Centella asiatica (L.) extract in increasing acetylcholine level in the heart of healthy wistar rats. Methods: This research is experimental in vivo with healthy wistar rats. The samples were divided into 5 groups, group 1 was the group without treatment, group 2 was the positive control group given Donepezil 3 mg / kgBW, group 3.4, and 5 were the groups given Centella asiatica (L.) extract 200, 400, and 800 mg / kgBW. After 21 days, each groups examined the heart acetylcholine level by ELISA. Data were analyzed statistically using independent t-test, paired t-test and further analysis using Bonferroni test. Results: There was no difference between (p> 0,05) between Centella asiatica (L.) extract with Donepezil in increasing acetylcholine level in the heart of healthy wistar rats, either 200, 400 or 800 mg / kgBW. However, there is a tendency to increase acetylcholine levels in 400mg / kgBW group. Conclusion: Therefore it can be concluded that no difference in the effectiveness between Centella asiatica (L.) with Donepezil in increasing acetylcholine level in the heart of healthy wistar rats.
