  • Green betel leaf (Piper Betle L.) contains essential oils that have antibacterial activity. The research aimed to analyse the effect volume variation of essential oils (Piper Betle L.) incorporated into HPMC (Hydroxy Propyl Methyl Cellulose) and chitosan patch on the physical properties and antibacterial activity against Staphylococcus aureus. Patch film was made by the solvent casting method with varying concentrations of essential oils, that were (110 mg/mL, 112 mg/mL, 114 mg/mL, 116 mg/mL, and 118 mg/mL). Physical properties and antibacterial activity of patch were tested, the result showed that the five formulas had variance of weight, thicknes and folding endurance appropriate with the standard, also good swelling index. Minimum inhibitory concentration of the patch was determined by observing the optical density ratio. Formula 4 contained 116 mg/mL essential oils of Piper Betle L. were elected as MIC because it could inhibited Staphylococcus aureus until 98,06%. Data of physical properties was analyzed using Anova variance statistical. The antibacterial compounds in essential oils that have incorporated to the patch were analyzed qualitatively using GC-MS method. Antibacterial compounds detected were eugenol, isoeugenol, eugenol acetate, cinnamaldehyde, and chavicol.
