  • The Flacourtia rukam plant has been used traditionally for the treatment of various diseases related to antioxidant and antibacterial activity. In this research, the antioxidant and antibacterial activity of Flacourtia rukam stem bark extract has been done. This research began with extraction by maceration and soxhlation using ethanol and ethyl acetate solvents. Each extract was determined percentage yield. The research continued with thin layer chromatography analysis on each type of extract. Water content on each type of extract was determined using the gravimetric method. Total phenolic and flavonoid levels were determined using a UV-Vis spectrophotometer. The antioxidant activity tested by using the DPPH method and antibacterial activity using the disk diffusion method. The results showed the highest value of percentage yield obtained in ethanol extract by maceration was 8.68%. In TLC analysis, it was found in the stain pattern between the same maceration and soxhlation method but different dark light patterns related to the quantity of compound components. The best water content was found in maceration extraction using ethyl acetate as 6.07%. Determination of total phenolic and flavonoid levels showed extracts of Flacourtia rukam stem bark with maceration ethanol had the highest total phenolic and flavonoid values (14.022 mg GAE/g and 11.458 mg QE/g) compared to other extracts. Determination of antioxidant activity obtained IC50 values in maceration and soxhlation ethanol extract of 185.13 mg/L, 196.92 mg/L and in maceration and soxhlation ethyl extract of 151.336 mg/L, 148.16 mg/L. Antibacterial activity showed that ethanol dan ethyl acetate soxhlation extract stem bark Flacourtia rukam had antibacterial activity against gram negative and ethanol soxhlation, ethyl maceration, ethyl soxhlation had antibacterial activity against gram positive at each 1000 mg/L with zone inhibition below 7 mm. Based on this data, it was concluded that stem bark extract of Flacourtia rukam was active as antioxidant and was weak as antibacterial.
