ISOLASI DAN IDENTIFIKASI BAKTERI ENDOFIT RUMPUT LAUT SPESIES Halimeda macroloba DARI PULAU MASPARI, OGAN KOMERING ILIR SUMATERA SELATAN
Daftar Isi:
- H. macroloba is one seaweed of the Chlorophyta (green algae) which produceds bioaktive compound. One of the efforts is making to explore of bioaktif compounds i.e. bacterial endophyte. This research aims to found and analyzed the amount, type and rate of growth of bacterial endophyte association H. macroloba from Maspari Island, South Sumatra. Procedures performed include growth, isolation, purification, staining Gram, biochemical test, identification and growth rate. Insolation is made with pour plate techniques and obtained 3 isolates pure with code isolates i.e HL2K (Veilonella sp.), HL2P (Yersinia sp.) and HL2SPK (Shigella sp). Isolates are obtained entirely negative Gram and has a characteristic i.e Veilonella sp. (cocus has yellow colonies), Yersinia sp. (basil has white colonies) and Shigella sp (basil has orange colonies). The largest number of bacteria isolates the bacteria Shigella sp., Yersinia sp., and Veilonella sp. in a row is 39 x 1010 (Cfu/ml), 12.7 1010 (Cfu/ml) and 271 x 1010 (Cfu/ml) and initial concentrations i.e., 79 x 104 (Cfu/ml), 184 x 104 (Cfu/ml) and 37 x 104. (Cfu/ml). The highest growth rate of any consecutive isolates 0.78 (Cfu/ml/hour), 0.95 (Cfu/ml/hour) and 1.49 (Cfu/ml/hour). The fastest generation time of any bacteria that is 13.17 hours (Shigella sp.), 14.05 hours (Veilonella SP.) and 6.99 hours (Yersinia sp.).
