  • Vernonia amygdalina Del. (asteraceae) is one of the plants which traditionally used as an antidiabetic. V. amygdalina old leaves and young leaves contains phenolic compounds, flavonoids, alkaloids, tannins, and saponins. This study aimed to determined the effectiveness of ethanol extract of V. amygdalina old leaves and young leaves in inhibiting α-glucosidase enzyme activity and determining total phenolic compound. The inhibition test has been done by measuring the absorbance at wavelenght 405 nm of p-nitrophenol which formed as a product from enzymatic reaction. The result showed IC50 of α-glucosidase activity ethanol extract of young leaves are 18.37 ppm and ethanol extract of old leaves are 43.05 ppm less than acarbose with 46.63 ppm. The total phenolic compound determined by spectrophotometry using Folin Ciocalteau. The total phenolic in ethanol extract of young leaves are 260 mgGAE/g and ethanol extract of old leaves are 196.5 mgGAE/g. Based on the result of study concluded that ethanol extract of young leaves have the bigger effect in inhibiting α-glucosidase than ethanol extract of old leaves or acarbose and have the highest total phenolic.
