PERBANDINGAN PENGGUNAAN METODE CTAB DAN KIT PADA ISOLASI DNA PADI VARIETAS LOKAL DI SUMATERA SELATAN

- Potential wetlands of South Sumatra such as tidal land is a major rice-producing areas. Local rice or land rice germplasm is a potential source of genes that control important traits in rice plant. Local varieties of rice have several deficiencies that cause farmers began to leave. The purpose of this study was to compare the results of the DNA isolation of local varieties of rice with different methods i.e. CTAB and KIT so it can determine the type of a method where appropriate for the isolation of DNA. The quality and quantity of DNA to high contaminant-free and can be used for other studies using PCR. This research was carried out in August 2015 until December 2015. This research was conducted in the laboratory of Microbiology and Genetics Laboratory, Department of Biology Biotechnology &, Faculty of Mathematics and Natural Sciences, University of Sriwijaya. This research includes the isolation of DNA using two methods i.e. CTAB and KIT, qualitative test with electrophoresis and quantitative test with UV Vis spectrophotometers. The measurement of the concentration of DNA with a UV Vis spectrophotometer is done at a wavelength of 260 nm. Purity of DNA solution can be calculated through the comparison of the A260 nm with A280 nm. The boundaries of the purity of the commonly used in molecular analysis on the A260/A280 ratio was 1.8 – 2.0. Next do some checking the quality of DNA gel electrophoresis results with a view to find out the level of purity of the DNA from the RNA and DNA integrity of contaminant results DNA isolation. The results of the DNA isolation of local varieties of rice leaves using CTAB method shows almost all the DNA bands generated has a bad quality and still smear. Quantitative test results using UV Vis spectrophotometer obtained still contained DNA that contaminated by phenols. The purity of the DNA on the range numbers 1-2,87. While the results of qualitative testing DNA electrophoresis local rice using KIT shows fragments of DNA or Ribbon that is still thin and smear. Test results obtained by UV Vis spectrophotometer using quantitative DNA with the purity of DNA on a range of 1-2,66. Thus between the two methods of DNA isolation used so produces DNA methods with better quality is CTAB method while quantity is KIT method.
