  • Infectious diseases is the type of disease most suffered by people in developing countries such as Indonesia. The microorganisms that cause infections are Escherichia coli and Staphylococcus aureus. This research aims to determine the antibacterial activity of fractions and active compound from Mahoni leaves (Swietenia mahagoni (L.) Jacq.) extract against Escherichia coli and Staphylococcus aureus. This research has been held from September to December 2016. The method in this research applied extraction with maceration method,fractionation with liquid fractionation method (FCC), antibacterial activity using agar diffusion method and isolation of active compound with the earth gravity column chromatography. Bacteria used in this research were Escherichia coli and Staphylococcus aureus. The result of this research showed that fraction of ethanol water and fraction of ethyl acetate is better inhibiting the growth of E. coli and S. aureus, compared with fraction of n-hexane it seen from the diameter of inhibition zone formed of each fraction. Minimum Inhibitory Concentration (MIC) from fraction of ethyl acetate on E. coli was 125 μg/ml and S. aureus was 500 μg/ml. The result purification of the active compound obtained E21 isolate estimated the alkaloid compound with Rf value of 0,66. Minimum Inhibitory Concentration (MIC) of active compounds on E. coli was 125 μg/ml and S. aureus was 250 μg/ml. The conclusion of this research was Mahoni leaves has antibacterial compound which able of inhibiting the growth of E. coli and S. aureus, thus Mahoni leaves extract contained antibacterial compound the alkaloid.
