  • Vitamin D is grouped in Biopharmaceutics Classification system (BCS) as class IV which means it has low solubility in water and low permeability. Low solubility and permeability will affect the drug's ability to be absorbed in the body, resulting in less therapeutic effect. Preparation of vitamin D into the form of submicro particles consisted polimer chitosan, sodium alginat, and cross linker calcium chloride using ionic gelation method. Calcium chloride varied concentration to 3 formulas produce EE percentage of 94.2% (20 μL); 87.9% (40 μL); 77.2% (100 μL). Based on result of EE percentage, the optimum formula of submicro particles chitosan sodium alginat vitamin D contained 20 μL calcium chloride. The result of submicro particles characterization such as diameter and particles distribution (poly dispersity inde/PDI) and potential zeta using Particle Size Analyzer (PSA) were 264.4 nm; 0.325; and 30.2 mV respectively. Morphologic particle determination on optimum formula using transmission electron microscope (TEM) showed spherical shape particle. Permeability of particles was observed through vitro diffusion testing using Franz diffusion cell. The result of diffusion test during 24 hours showed the submicro particles was 10.775% compared to pure vitamin D solution was 9.381%.
