- Trichophyton is the most common causes of dermatomycosis. Ketoconazole can be used as a treatment of dermatomycosis. Ketoconazole active ingredient to class II of Biopharmaceutics Classification System (BCS) which shows that the solubility is low in water and has good permeability. Low solubility affects the bioavailability of a drug and its absorption on the body. The preparation of ketoconazole into submicro particles consisted of chitosan polymer, sodium alginate and cross linker CaCl2 by ionic gelation method. The calcium chloride concentration varied on the three formulas yielded % entrapment efficiency (% EE) of 90.77% (20 μL); 92.15% (40 μL); 94.5% (100 μL). Based on the results of percent EE, the optimum formula of submicro particles chitosan sodium alginate ketoconazole with the concentration of calsium chloride is 100 μL. The results of particle characterization such as diameter and particle distribution (Poly Dispersity Index / PDI), and zeta potential using particle size analyzer (PSA) tool in optimum formula is 287.3 nm; 0.325; And 24.1 mV. The observation of particle morphology using transmission electron microscope (TEM) tool to the formulas of chitosan submicro particles sodium alginate ketoconazole shows particles with spheric shape. Particle permeability was observed through in vitro diffusion testing using Franz diffusion cells. The 24-hour diffusion test results showed diffused percentage on submicro particles is 4.241%, compared with a pure ketoconazole solution is 2.664% and ketoconazole dosage from market is 3.102%. The effectiveness of the drug was observed through in vivo testing using animal test infected with Trichophyton. Submicro particle show the greatest decrease of lesions in the target when compared with pure ketoconazole and commercial ketoconazole.
