OPTIMASI WAKTU SONIKASI, KADAR POLY(LACTIC-CO-GLYCOLIC ACID), DAN POLY(VINYL ALCOHOL) PADA FORMULASI MIKROPARTIKEL FIKOSIANIN DENGAN DESAIN BOX-BEHNKEN

- A research on the optimization of the influence of sonication time, Poly(Lactic-co-Glycolic Acid) (PLGA) concentration, and Poly(Vinyl Alcohol) (PVA) formulation of phycocyanin microparticle with Box-Behnken design has been done. Phycocyanin is a greenish-blue pigment derived from Spirulina platensis which has antioxidant activity and prevents premature aging. Phycocyanin preparation into microparticle dosage form using polymer PLGA and stabilizer PVA with double emulsion solvent evaporation method aims to improve the stability of phycocyanin. The optimization of phycocyanin microparticle with Box-Behnken design in Design-Expert®10 program is useful to see the variation effect of PLGA concentration (0,1-2%), PVA concentration (1-2%), and sonication time (5-30 minutes) toward the test responses such as percent efficiency encapsulation (%EE), viscosity, and pH. The optimum formula is obtained from the values of the test responses of 15 basic formulas. The optimum formula compositions obtained are PLGA concentration 0.1%, PVA concentration 1.22%, and sonication time 5 minutes, with the test response values are %EE of 92.235%, viscosity of 0,940 cP, and pH of 7. The freeze drying result of optimum formula reaches a good percent process yield (%PY) which of 59.373%. The analysis of average diameter, poly dispersity index (PDI), and zeta potential using particle size analyzer (PSA) are 1.565 μm, 0.541, and -2.2 mV. The analysis of particle morphology using scanning electron microscopy (SEM) showed an inter-particle agglomeration. The microparticle dosage forms manage to increase the phycocyanin stability indicated by concentration percentage decrease. The stability test is conducted on microparticle dosage forms and free phycocyanin. The results of percent concentration decrease obtained from thermodynamic stability test for 2 cycles are each 23.571% and 62.544%, while the results of pH stability test at pH 6,8 are 20.941% and 25.806%. The analysis FTIR (Fourier Transform Infrared) shows no chemical interaction in phycocyanin and polymer PLGA-PVA on the results of the infrared spectrum of phycocyanin microparticle.
