PENGARUH ZAT PENGATUR TUMBUH DAN LAMA SIMPAN BENIH TERHADAP PERTUMBUHAN ZIGOTIK EMBRIO BENIH KELAPA SAWIT (Elaeis guineensis Jacq. Var. TENERA) SECARA IN VITRO

- This research was conducted to determine whether growth regulators (PGR) and long seed storage affect the Growth of Zygotic Palm Oil Embryo (Elaeis guineensis Jacq. Tenera) In Vitro. The study was conducted at the Tissue Culture Laboratory of PT. Sampoerna Agro Tbk, Km 10, Palembang city, South Sumatra Province in July 2018 until completion. The study used the Tenera palm oil embryo. This study uses two treatment factors, namely culture media and seed shelf life. Media treatment M1 = 0.1 NAA + 0.1 BAP + 0.1 GA3 + 2000 Active Charcoal, M2 = 0.1 NAA + 0.5 BAP + 0.1 GA3 + 2000 Active Charcoal, M3 = 0.1 NAA + 1 BAP + 0.1 GA3 + 2000 Active Charcoal, M4 = No ZPT + No Active Charcoal. Treatment of long seed storage of U1 = Seed save age <1 year, U2 = Seed save 2 year age <3 years, U3 = Seed save age> 3 years. This research used RCD and continued with LSD test of 5% level. The results showed that the use of growth regulators and longevity of seed saving significantly affected the growth and development of zygotic palm oil embryos in vitro. in general, treatment media 0.1 NAA + 1 BAP + 0.1 GA3 + 2000 Active Charcoal and seeds with a long shelf life of 2 years <age <3 years have the best results.
