- Free radicals are one of the causes of degenerative diseases that can be inhibited by antioxidants. The source of natural antioxidants comes from Kersen leaf since it has secondary metabolites in the form of flavonoids. The flavonoid compounds contained in ethanol extract of Kersen leaf in vitro have strong antioxidant power with IC50 values of 79,96 ± 0,91 μg/mL. The purpose of this study was to determine the antioxidant effects of Kersen leaf ethanol extract in vivo using the TBARs method by measuring plasma MDA levels of male rats induced by carbon tetrachloride (CCl4), the measurement was based on the formation of the MDA-TBA complex. This study used 30 male white rats Wistar strain which were divided into 6 groups, namely the normal group (Na CMC 1%), negative (Na CMC 1%), positive (Vitamin C 1,98 mg/200 gBB), and treatment 1, 2 and 3 (ethanol extract of kersen leaves doses of 150, 300, and 600 mg/kgBB) were given orally for 6 days while the 7th day of rats were induced with CCl4 except for normal control. The results of the study using the TBARs method showed that the three doses of extract had an effect as antioxidants because they could reduce the plasma MDA levels of mice by 1,565 ± 0,316 nmol/mL, 0,809 ± 0,330 nmol/ mL, and 0,548 ± 0,169 nmol/mL. Ethanol extract of Kersen leaf has an antioxidant effect and the best effect as an antioxidant is a dose of 600 mg/kg because it has an antioxidant effect that is almost the same as vitamin C (0,395 ± 0,199 nmol/mL).
