  • A research about Optimization of Early Detection of Potato Virus Y (PVY) in Potato Solanum tuberosum L. varieties Kalosi by ELISA Technique (Enzyme linked immunosorbent assay) was done. The purpose of this research was to determine the level of sensitivity of the combination of antibody and antigen dilutions with ELISA technique in detecting the virus PVY in potato plants. The research was carried out in the Laboratory of Agricultural Biotechnology, Research Center of Hasanuddin University, Makassar. Sample was tested using extracts of potato tuber varieties kalosi Generation 2. Leaf extract of infected potato was used as positive control, and buffer extract as a negative control. In the early stages of a process of optimization of the ELISA reagents through the process of sample extraction positive control, dilutions of antigen and antibodies was used. It was aimed to determine the minimum level of dilution that can still be used to detect the virus PVY. Observation measured by Elisa Reader that operated in wavelength 405 nm. Positive result signed if tested samples has average point more than 2x point of negative sample. The results showed that the optimization of some level of dilution suggests that the level of antigen dilutions up to 10-4 could still detect the presence of the virus PVY and the maximum level of antibody dilution is 1:1200. Testing the results against the potato tuber varieties kalosi on a combination of antibodies (10-3) with antigen (10-3) did not reveal any positive reaction to the virus PVY.
