- Acute Lymphoblastic Leukemia (ALL) is a type of leukemia that is often found in children with a variety of molecular patomechanisms. The process of apoptosis is one that plays an important role in ALL patomechanism and it is influenced by a variety of proteins that are pro-apoptotic and antiapoptotic. This study was conducted to determine the role of these pro- and antiapoptosis proteins ratios in the apoptotic susceptibility of primary culture cells ALL measured by Annexin V, propidium iodide and mitotracker RedCMOS. FACS Calibur Flowcytometry was used to determine the immunophenotype and the expression of various proapoptotic proteins such as caspase-3, Bax, p53 and antiapoptotic proteins such as survivin, Bcl-2 and MDM2. The same method was also used to measure apoptosis markers such as annexin-v, propidium iodide and mitotracker RedCMOS. As for determining the role of various proteins to apoptosis, the researchers analyzed the comparative figures of the intensity of expression of each protein and compared with the percentage of cells undergoing apoptosis. On the four (4) primary ALL cells cultured isolates, the researchers obtain the intensity of expression: Bax/Bcl-2, p53/MDM2 different, whereas the comparative figures of caspase-3/survivin was relatively same. Based on this research, it can be concluded, thatthe vulnerability of apoptosis in vitro correlates strongly to influence the results of several apoptotic proteins such as survivin/caspase-3,bax/bcl-2 and p53/MDM2. Comparative figures of p53/MDM2 is the most influential factor on apoptosis resistance in ALL children compared with other relevant proteins
