  • ABSTRACT The optimum production time was 25 for Bacillus BE-1 with collagenase activity of 0.086 U/ml. When collagen was added to the Modified LB media (LB + Collagen) different responses was obtained. In this case, we reduced the nutrient content (carbon and nitrogen sources) of LB, such that the bacteria was encouraged to utilize more collagen substrate added into the media. After 15 hours in LB + collagen media, the activity of collagenase of Bacillus BE-1 was increased from 0.086 U/ml to 0.094 U/ml. The Substrate spesificities in LB media showed collagenase from Bacillus BE-1 was more active towards fibrin compare with the other 3 protein substrates (casein, collagen and gelatin). Addition of collagen in the media changed the substrate spesificities of the Collagenase. Collagenase from Bacillus BE-1 became more active towards collagen eventhough casein remained the best substrate. Key Words: Collagenase, Bacillus BE-1, spesificities
