- The objective of this green house experiment was to determine the most virulent among 10 B. bassiana isolates against P. xylostella larvae. The tested isolates were originally obtained from several insects, i.e. Nilaparvata lugens, Plutella xylostella, Chrysodeixis chalcites, Hypothenemus hampei, Conopomorpha cramerella, Thrips tabaci. Conidia from slant culture of GYA added chitin from small mole criket diluted to get 101, 103, 105, and 107 conidia per ml. The third instar of P. xylostella were put on mustard leaves and then B. bassiana isolates were inoculated by spraying them on the leaves. This bioassay was carried out to determine median lethal time (LT50) of the isolates, and mortalities of larvae and pupae. Three lowest LT50 at 1 x 107 conidia per ml of C. chalcites (KBC), C. cramerella (S100), and C. chalcites (PD2) isolates were 17.1, 20.6, and 27.4 hours, respectively. At 1 x 1 x 107 conidia per ml, all isolates were able to kill more than 60% larval stages after 48 hour application.
