  • ABSTRAK PENGARUH PEMBERIAN VITAMIN C TERHADAP JUMLAH SEL PURKINJE CEREBELLUM TIKUS PUTIH (Rattus norvegicus) JANTAN GALUR Sprague Dawley YANG DIINDUKSI MONOSODIUM GLUTAMAT Oleh ISMALIA QANIT Latar belakang: Konsumi MSG yang berlebihan dapat menyebabkan gangguan pada otak, salah satunya yaitu cerebellum. Glutamat yang berlebihan menghasilkan eksitotoksistas endogen yang terbukti memicu terjadinya stress oksidatif sehingga menyebabkan penurunan kepadatan jumlah sel purkinje di cerebellum. Antioksidan pada vitamin C diharapkan dapat meningkatkan jumlah sel purkinje cerebellum. Tujuan: Tujuan dari penelitian ini adalah untuk mengetahui pengaruh vitamin C terhadap jumlah sel purkinje cerebellum tikus putih (Rattus norvegicus) jantan galur Sprague dawley yang diinduksi MSG. Metode: Penelitian ini adalah studi eksperimental dengan rancangan Post Test Only Control Group Design. Penelitian dilakukan selama 21 hari. Sampel berjumlah 30 ekor, dibagi dalam 5 kelompok, yaitu K- (aquadest), K+ (MSG 4mg/grBB), P1 (MSG 4mg/grBB dan vitamin C 100 mg/kgBB), P2 (MSG 4mg/grBB dan vitamin C 200 mg/kgBB), dan P3 (MSG 4mg/grBB dan vitamin C 400 mg/kgBB). Kemudian dilakukan penghitungan jumlah sel purkinje cerebellum dengan metode touch count menggunakan mikroskop cahaya olympus pada perbesaran 400x dalam 5 lapang pandang. Hasil: Hasil rerata jumlah sel purkinje secara berurutan setiap kelompok (K-), K(+), P1, P2 dan P3 adalah 54,2; 34,8; 49,6; 53,6; 43,6. Hasil Uji One Way Annova didapatkan p=0,005. Uji Post Hoc LSD terhadap jumlah sel purkinje menunjukkan perbedaan yang bermakna antara K- dengan K+ (p=0,001), antara P1, P2 dengan K+ (p<0,05). Simpulan: Terdapat pengaruh pemberian vitamin C dosis 100 dan 200mg/kgBB/hari terhadap jumlah sel purkinje cerebellum tikus putih (Rattus novergicus) jantan galur Sprague dawley yang diinduksi MSG. Kata Kunci: Cerebellum, Monosodium glutamat, Vitamin C ABSTRACT THE EFFECT OF VITAMIN C TO THE NUMBER OF PURKINJE CEREBELLUM CELLS ON MALE ALBINO RAT (Rattus norvegicus) STRAIN Sprague Dawley INDUCED BY MONOSODIUM GLUTAMAT By ISMALIA QANIT Background: Consumption of MSG in excessive way can increase risk for brain, one of it‟s part is cerebellum. An Excessive glutamate can produce endogenous excitotoxicity which is proven to trigger oxidative stress, then the number of purkinje cells will decrease. Antioxidant in vitamin C is expected to increase the number of purkinje cerebellum cells. Objective: To know the effect of vitamin C to the number of purkinje cerebellum cells on male albino rat (Rattus norvegicus) strain Sprague dawley induced by MSG. Method: This is an experimental study with Post Test Only Control Group Design. This experimental study carried out for 21 days. The samples were 30 rats divided into 5 groups which are, K- (aquadest), K+ (given MSG 4 gr/kgBB), P1 (MSG 4 gr/kgBB and vitamin C 100 mg/kgBB), P2 (MSG 4 gr/kgBB and vitamin C 200 mg/kgBB) and P3 (MSG 4 gr/kgBB and vitamin C 400 mg/kgBB). Next, calculate the number of purkinje cerebellum cells by the touch count method using the Olympus light microscope with 400x magnification in 5 visual fields. Results: The mean number of purkinje cerebellum cells results in sequence for each group (K-), K(+), P1, P2 dan P3 were 54,2; 34,8; 49,6; 53,6; 43,6. The results of One Way Annova analysis p=0,005. Post Hoc LSD test shown significant diffrence between K- with K+ (p=0.001), between P1, P2 with K+ (p<0,05). Conculsion. There is an effect of vitamin C 100 and 200 mg/kgBB/days to the number of purkinje cerebellum cells on male albino rat induced by MSG. Keywords: Cerebellum, Monosodium, Vitamin C
