- ABSTRACT The Effect of Moringa Oleifera Leaf Extract on Malondialdehyde (MDA) Eye Lens in Male Albino Rats (Rattus novergicus) Sprague dawley Strain Induced By Sodium Selenite By RIZKA DWI FEBRIYANTI Background: Cataract is a condition of turbidity that occurs in the lens of the eye caused by hydration of lens fluid, protein denaturation or the result of both which are still the main problems of blindness in various countries including Indonesia. Indonesia ranks first with the number of cataract sufferers in Southeast Asia. Cataract formation can be triggered by an increase in intermediate reactive oxygen (ROI) which exceeds the endogenous antioxidant defenses resulting in an excess of free radicals. Antioxidants can prevent the formation of free radicals. Moringa oleifera leaves contain antioxidants that can stop free radicals. Methods: This experimental study was carried out for 14 days. 30 male Sprague dawley rats were divided into 5 different groups: K1 was not treated, K2, P1, P2, P3 were induced by sodium selenite wit h a dose of 15 μmol / kgBB subcutaneously; P1, P2, P3 were given Moringa leaf extract at a dose of 72 mg / kgBB (P1), 144 mg / kgBB (P2), 288 mg / kgBB (P3). Malondialdehyde levels were measured by the TBARS test method (Thiobarbituric Acid Reactive Substances). Data were analyzed by one way ANOVA test. Results: Based on the one way ANOVA test, Moringa oleifera leaf extract was used to reduce malondialdehyde levels in the eyes of male white rats induced by sodium selenite (p = 0,000). The mean malondialdehyde level was 5.72 ± 0.41 (K1); 8.23 ± 0.51 (K2); 7.76 ± 0.51 (P1); 7.18 ± 0.47 (P2); 6.25 ± 0.40 (P3). In the post hoc bonferroni test, different levels of doses of 144 mg / kgBB and 288 mg / kgBB had significant differences with the treatment group that was only induced by sodium selenite (K2). Conclusion: There is an effect of Moringa oleifera leaf extract on the levels of malondialdehyde lens in the eyes of male albino rats induced by sodium selenite. Decreasing levels of malondialdehyde are more significant with increasing doses. Keywords: Malondialdehyde, Moringa oleifera, sodium selenite. ABSTRAK PENGARUH PEMBERIAN EKSTRAK DAUN KELOR (Moringa oleifera) TERHADAP KADAR MALONDIALDEHID (MDA) LENSA MATA TIKUS PUTIH (Rattus novergicus) JANTAN GALUR Sprague dawley YANG DIINDUKSI SODIUM SELENITE Oleh RIZKA DWI FEBRIYANTI Latar Belakang: Katarak merupakan keadaan kekeruhan yang terjadi pada lensa mata yang disebabkan oleh hidrasi cairan lensa, denaturasi protein atau akibat keduanya yang masih menjadi masalah utama kebutaan di berbagai negara termasuk Indonesia. Indonesia menempati peringkat pertama dengan jumlah penderita katarak di Asia Tenggara. Pembentukan katarak dapat dipicu oleh peningkatan oksigen reaktif intermediate (ROI) yang melebihi pertahanan antioksidan endogen sehingga terjadi kelebihan radikal bebas. Antioksidan mampu mencegah pembentukan radikal bebas. Daun kelor (Moringa oleifera) memiliki kandungan antioksidan yang mampu menghentikan radikal bebas. Metode: Penelitian eksperimental ini dilakukan selama 14 hari. Sebanyak 30 ekor tikus putih jantan galur Sprague dawley dibagi kedalam 5 kelompok yang berbeda: K1 tidak diberi perlakuan, K2, P1, P2, P3 diinduksi sodium selenite dosis 15 μmol/kgBB secara subcutan; P1, P2, P3 diberikan ekstrak daun kelor dengan dosis 72 mg/kgBB (P1), 144 mg/kgBB (P2), 288 mg/kgBB (P3). Kadar malondialdehid diukur dengan metode TBARS test (Thiobarbituric Acid Reactive Substances). Data dianalisis dengan uji One Way ANOVA. Hasil Penelitian: Berdasarkan uji one way ANOVA pemberian ekstrak daun kelor (Moringa oleifera) berpengaruh untuk menurunkan kadar malondialdehid lensa mata tikus putih jantan yang diinduksi sodium selenite (p=0,000). Rerata kadar MDA adalah 5,72 ± 0,41(K1); 8,23 ± 0,51 (K2); 7,76 ± 0,51 (P1); 7,18 ± 0,47 (P2); 6,25 ± 0,40 (P3). Pada uji post hoc bonferroni, perbedaan dosis bertingkat yaitu dosis 144 mg/kgBB dan 288 mg/kgBB memiliki perbedaan yang bermakna dengan kelompok perlakuan yang hanya diinduksi sodium selenite (K2). Kesimpulan: Terdapat pengaruh pemberian ekstrak daun kelor (Moringa oleifera) terhadap kadar malondialdehid lensa mata tikus putih jantan yang diinduksi sodium selenite. Penurunan kadar malondialdehid semakin bermakna seiring dengan penambahan dosis pemberian. Kata kunci: Malondialdehid, Moringa oleifera, sodium selenite.
