  • Metode pulping formacell adalah salah satu cara pengolahan tandan kosong kelapa sawit menjadi pulp dan menghasilkan lindi hitam sebagai limbah. Lindi hitam memiliki dampak negatif terhadap lingkungan jika tidak dilakukan pengelolaan dengan baik maupun pengolahan lebih lanjut. Lindi hitam diketahui memiliki kandungan lignin sebesar 46%. Lignin merupakan komponen lignoselulosa yang diketahui memiliki efek aktivtitas prebiotik dan antimikroba dikarenakan sifatnya yang sulit dicerna dan terdiri dari komponen fenolpropanoid. Penelitian sebelumnya menunjukkan bahwa lignin hasil pemurnian menunjukkan aktivitas prebiotik dan diduga memiliki aktivitas antimikroba pada rumen hewan ternak. Penelitian ini bertujuan untuk melihat aktivitas antimikroba dan aktivitas prebiotik pada fraksi lignin hasil pemurnian. Teknik identifikasi fraksi lignin dilakukan dengan menggunakan plat Kromatografi Lapis Tipis (KLT) dan Gas Chromatography-Mass Spectrometri (GC-MS). Uji aktivitas antimikroba menggunakan metode difusi agar cakram terhadap Escherichia coli dan uji aktivitas prebiotik dilakukan menggunakan perhitungan total bakteri pada pertumbuhan Lactobacillus casei. Metode peneltian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan tiga kali ulangan. Faktor tunggal perlakuan adalah konsentrasi fraksi lignin yaitu 0%, 2,5%, 5%, 7,5%, 10%, 12,5%, dan 15%. Rendemen fraksi CHCl3, 3%MeOH:CHCl3, 20%MeOH:CHCl3, dan MeOH secara berurutan sebesar yaitu 10,68%, 6,34%, 11.38% dan 44,85%. Fraksi 3% MeOH:CHCl3 diidentifikasi mengandung senyawa benzaldehyde, 4- hydroxy-3,5-dimethoxy, 1-methtylbutyl hexadecanoate, oleic acid, dan di-2- ethylhexyl phthalate. Fraksi 3%MeOH:CHCl3 pada konsentrasi 15% menghasilkan zona hambat sebesar 6,33 mm terhadap E. coli dan menunjukkan aktivitas sebagai prebiotik terhadap L. casei sebesar 6,1 x 102 koloni/mL. Kata kunci: antimikroba, lignin, lindi hitam, prebiotik, tandan kosong kelapa sawit ABSTRACT The formacell is pulping method of processing oil palm empty fruit bunches into pulp and producing black liquor as a process waste. Black liquor has a negative impact on the environment if it is not carried out properly management or further processing. Black liquor is known to have a lignin content of 46%. Lignin is a lignocellulosic compenent that is known to have an active effect on prebiotics and antimicrobials due to its hard-to-digestive properties and consists of the phenolpropanoid component. Previous research showed that purified lignin showed prebiotic activity and was thought to have antimicrobial activity in the rumen of livestock. This study aims to look at the antimicrobial activity and prebiotic activity on purified lignin. The technique of identifying lignin monomers was carried out using Thin Layer Chromatography (TLC) plates and Gas Chromatography-Mass Spectrometry (GC-MS). Antimicrobial activity test using disk difusion agar method against Escherichia coli and prebiotic activity tests were carried out using calculation of total plate count on the growth Lactobacillus casei. The research method used was a Completely Randomized Design (CRD) with three replications. The single factor of treatment was the concentration of lignin fraction which were 0%, 2.5%, 5%, 7.5%, 10%, 12.5%, and 15%. The yield of CHCl3, 3%MeOH:CHCl3, 20%MeOH:CHCl3, and MeOH fractions respectively were 10.68%, 6.34%, 11.38% and 44.85%. The 3%MeOH:CHCl3 fraction was identified to contain benzaldehyde, 4-hydroxy-3,5- dimethoxy, 1-methyllbutyl hexadecanoate, oleic acid, and di-2-ethylhexyl phthalate. The 3%MeOH:CHCl3 fraction at concentration 15% has shown inhibition zone of 6.33 mm against E. coli and activity as prebiotic for L. casei with total microbes of 6.1 x 102 colony/mL.
