  • This research aims to determine the effect of kinetin in various concentration on culture shoot induction of Tobacco. This research was conducted in October to December 2018 at Tissue Culture Laboratory of State Polytecnic of Jember. This research was using Completely Randomized Design non factorial with treatment concentration of kinetin in MS media that consisted 6 levels, there were 0 ppm (P1), 1 ppm (P2), 2 ppm (P3), 3 ppm (P4), 4 ppm (P5) and 5 ppm (P6). Data were analyzed using Analysis of Variance (ANOVA) and if significantly different continued with Duncan Multiple Range Test (DMRT) 5%.. The results of this research show that Kinetin giving is significant effect on cocentration of 3-5 ppm, with the best concentrations on early budding are 4 ppm and 5 ppm of 12,8 – 13,2 days after inoculation, explant sprout 100%, average value height of shoot is 1,15 cm and average value of number of shoot is 108.
  • Penelitian ini bertujuan untuk menentukan pengaruh berbagai konsentrasi kinetin pada kultur tunas induksi Tembakau. Penelitian ini dilakukan mulai bulan Oktober sampai dengan Desember 2018, di Laboratorium Kultur Jaringan Tanaman, Politeknik Negeri Jember. Rancangan penelitian menggunakan Rancangan Acak Lengkap Non Faktorial dengan faktor variasi konsentrasi Kinetin pada media MS yang terdiri dari 6 taraf perlakuan yaitu 0 ppm (P1), 1 ppm (P2), 2 ppm (P3), 3 ppm (P4), 4 ppm (P5), 5 ppm (P6). Data hasil pengamatan penelitian dianalisis menggunakan Sidik Ragam (ANOVA), apabila berbeda nyata maka dapat diuji lanjut dengan menggunakan Duncan Multiple Range test (DMRT) taraf 5 %. Hasil penelitian menunjukkan bahwa pemberian Kinetin berpengaruh nyata pada konsentrasi 3-5 ppm, dengan konsentrasi terbaik untuk kedinian tunas pada perlakuan 4 ppm dan 5 ppm yaitu 12.8 hari – 13.2 hari setelah inokulasi (HSI), eksplan bertunas 100%, tingkat kontaminasi 0%, rerata tinggi tunas 1.15 cm, dan rerata jumlah tunas 108 tunas.
