- The soybean can tie the nitrogen (N2) on the atmosphere through the activity of bacterium which is able to tie the nitrogen called Bradyrhizobium japonicum. This bacterium is formed inside the plant root called nodul or the small root. This small root of soybean is generally able to tie the nitrogen from the air aged 10-12 days after planting, which depends on the soil situation and its temperature (adisarwanto, 2005).The rhizobium can tie the nitrogen from the air and change it into nitrogen which can be used for the sake of soybean growth. If this seed divided into two, this small root will be red or pink. Those colored green or brown is not effective to tie the nitrogen (Pitojo, 2003). This study is undergone in Bendungan Sengguruh of perm Jasa Tirta area I in Sengguruh village of Kepanjen district of malang regency and in Biotech laboratory of UMM on April 24th 2007 up to August 2007. The equipments used in the field study are: hoe, sickle, measurement line, plastic rope, paper, plastic, “gejik” ( a tool to make a hole in soil). The materials used in the laboratory are : LAFC (laminar Air Flow Cabinet), Bunsen, Matches, autoclave, incubator, analytic scale, gas stove, microwave, Petridis, ose needle, beaker glass, rlenmeyer, bottle, spatula. The method used is planting five different varieties of soybean in which they are planted under the pole of mengkudu growing herd plant. The isolation process is done after taking the small roots from the soybean plant. The result of this study shows that we can get the best four different kinds of Bradyrhyzobium japonicum in which the have the best growth speed in peptone media put in the insulation temperature of 28 degree Celsius in the incubator grown in seven days. From the result of this study, we get some characteristics of Bradyrhyzobium japonicum which is able to have the mutualism symbioses with the soybean plant, it has the negative salt characteristics, in the form of bacillus, has a quick growth speed, the color of the colony ranges from white, yellow, and brown, the colony thickness ranges from the thinnest up the thickest which has the salivary characteristics up the least salivary, and it has the various edge of colony from circle up to branched one.
