- Background : Salicylic acid cannot be used orally so it has a derivative use, in order to reduce headache, muscle pain, but less effective for reducing toothache, menstrual pain, and cancer. In order to increase its analgesque effect from one of the descendants of salicylic acid as acetysalysalyylic, a structural modification was made by 5-chlorosalicylic acid synthesis with octanoyl chloride reactions through esterification. Purpose : O-(octanoyl)-5-chlorosalylic acid compounds can be produced from synthesize a 5-chlorosalicylic acid structure with octanoyl chloride using an esterification reaction from asil halide. An o-(octanoyl)-5-chlorosalicylic acid compound works asa better analgesic if compared with asetosal in a male's white spout (mus musculus). Research and Method : The synthesis of the o-(octanoyl)-5-chlorosalicylic acid compound uses the esterification of halide asyl which is 5-chlorosalicylic acid and octanoyl chloride. Some tests of purity included soft spots and phlesions with a different polychlorol acetone : methanol (6:4), chloroform: ethanol (9:1), and acetone: ethanol (5:1). Further testing is structural identification with uv-vis spectrophotometer, ir spectrophotometer, and spectrophotometer 1H-NMR. Analgesic activity tests using a corresponding thing test on the spark. Result and Conclusion : The result obtained from the synthesis was a distinctive, crusty, brownish crystal. And get 24,81% of the compounds produced, melting points gained an average of 130oC. In the result of the claw, found 1 stain using the elude; Aseton: methanol (6:4), ethanol: chloroform (9:1), aseton: ethanol (5:5). This study produced O-(octanoyl)-5-chlorosalicylic acid compounds. Qualitative analysis using uv-vis spectrophotometer, infrared and spectrometer 1H-NMR. Analgesic effects from the compound are as follows; 43.36% by 25 mg/kgBB, 46.69 by 50 mg/kgBB, and 60.67 % by 100 mg/kgBB. This notes that O-(octanoyl)-5-chlorosalicylic acid has analgesic activity and is greater than asetosal with an ED50 = 45.40 milligrams/kgBB.
