  • Natural dyes from different plant extracts are often done, but not all plants can be absorbed by preparations such as mitosis preparate. Secang (Caesalpinia sappan) in addition to being used as a fabric dye, food coloring, it can be used to dye the cell nuclei in the histotechnical tissue. Unfortunately, information about this is not accurate and thus requires an examination of optimum concentration of quality preparate. The purpose of this study is to find out the different concentrations of secang (Caesalpinia sappan) extracts to the quality of the Allium cepa mitosis staining preparation. This kind of research is a real experiment with the posstest research design. The amount of samples in this research is as much as 25 sample of Allium cepa mitosis preparations with 5 group treatment and 5 replications. The preparations are assessed based on the quality which includes the clarity of the preparations and the color contrast in the preparations. The data analysis technique used were Kruskal-Wallis and Mann-Whitney. The result showed that there was a significant difference in the concentration of secang (Caesalpinia sappan) extracts on the quality of the Allium cepa mitosis staining which was shown had a probability value of 0.002<0.05, so it did not have a significant difference with safranin coloring. The concentration of 80% is the best concentration of a cup of wood extract (Caesalpinia sappan) as a natural dye with the average score of a quality assessment of 3,9 which is based on the clarityof the preparation and the contrast of colors.
