  • The availability of preparations for the object in observing the structure and function of plant tissue is still lacking and not fully used in the learning procces. Making preparation requires staining to clarify and sharpen part of tissue and cells. The synthetic dyes is use to coloring tissues which has a relative expensive price and a slow absorption of color in the tissue, so an alternative dye is needed for coloring the preparation. Beetroot (Beta vulgaris L.) is an alternative natural coloring for the staining of section preparations of purslane plant (Portulaca oleraceae L.). The purpose of this study was to determine the effect of various concentrations of beetroot extract (Beta vulgaris L.) as natural dyes on the quality of the preparations of stems of purslane plants (Protulaca oleraceae L.). The type of research used is the actual experiment (The Experimental Research). The concentration used in this study is 0%, 15%, 30%, 45%, 60%, 75%, and 90%) and using safranin coloring. This study consisted of 24 samples of stump section preparations of purslane plants (Portulaca oleraceae L.). Evaluation of preparations based on the quality of preparaions includes clarity of preparations, color contrast and durability of preparations. The analysis technique used is Kruskall Wallis, Mann-Whitney and Friedman. The result showed was an influence of various concentration of beetroot extract (Beta vulgaris L.) as a natural dye on the quality of preparations of stems of purslane plants (Portulaca oleraceae L.) and preparations that have an effectice effect on qualityare extract of Beetroot (Beta vulgaris L.) 60% which have an average of 2,17 which isn’t much different form the safranin treatment having a score of 2,43.
