  • Potatoes (Solanum tuberosum L.) is one of the vegetable crops source of vitamins, mineral salts of wet tubers. BPS data in 2016 shows the number of potatoes production in 2016 reached 1,21 million tons, has decreased compared to 2015 which was 9,54 percent caused by the decrease in harvested area. The result of low commodities. The result of low commodities can be influenced by the availability of seeds by developing quality propagul potatoes are insufficient and the distribution of seeds are not qualified. Tissue culture biotechnology can produce seeds in large quantities and uniformly in a relatively short time and free of disease. Propagation of potato seedlings using tissue culture can use MS media. MS media has a quite expensive price and complicated manufacturing media requires the substitution of more economical materials such as leaf fertilizers that contain micro and micro elements. The success of tissue culture aside from growing media and must should involve plant growth regulator (ZPT) factors, one of which is BAP. This study aims to discusses the effect of leaf fertilizer concentration as an alternative medium for MS in in vitro culture of potatoes. The study was conducted in December 2018 - May 2019 in the Network Culture Laboratory of the Center for Biotechnology Development, Muhammadiyah University of Malang. The experiment was conducted using factorial randomized block design (RBD) in 3 replications, factor 1 was MS media concentration (1, 3⁄4, 2/4, 1⁄4) while factor 2 was leaf fertilizer concentration (0, 1, 2, 3 g / l ). The limitation of variable consisting of time appear shoots (HSI), high planlet (cm), number of leaves , number of segments , number of roots , number of tubers , chlorophyll levels, wet weight, dry weight, and plant growth. Based on the results of the study it can be concluded that MS media + Qiuvita leaf fertilizer 0 g / l (M0Q0) showed the best results on the growth of potato plantlets of Granola Kembang varieties on the time parameters appeared buds, root number, chlorophyll content and total chlorophyll. Full MS media treatment (MS) showed the best results on plantlet height, number of leaves, the best number of segments, and very hardened vigorous scoring at 42 HSI. The treatment of 0 g / l leaf fertilizer showed the best results on plantlet height, number of leaves, and number of segments while the concentration of 1 g / l also provided the best average number of leaves and number of segments at 42 HSI.
