  • Background: Piper betle L. is one of the plants that has antibacterial activity in Gram positive or negative gram bacteria. Staphylococcus aureus is the bacteria that may cause some disease such as skin diseases, bones, and pneumonia. Objective: This study aimed to investigate antibacterial activity Piper betle L. ethyl acetate fraction through the growth of Staphylococcus aureus in vitro and to find out the secondary metabolit in it. Method: Fraction of ethyl acetate piper betle L. was obtained by applying fraksinasi method. Residue powder of n-hexan macerare result was re-macerate with ethyl acetate solvent. The secondary metabolit was identified by using Thin Layer Chromatography (TLC) method and anti-bacteriae activity test was conducted by using disc diffusion method. Result and Conclusion: Secondary metabolit identification result found that fraction of etil asetat piper betle L contained polyphenols, terpenoid, and anthraquinone. Moreover, anti-bacterial activity test result Piper betle L. ethyl acetate fraction showed that was antibacterial activity in concentration 250 mg/mL (12,5 mg/disc) has diameter inhibition zone 15,83 mm; concentration 500 mg/mL (25 mg/disc) has diameter inhibition zone 16,10 mm; and concentration 750 mg/mL (37,5 mg/disc) has diameter inhibition zone 16,65 mm. The conclusion in this research antibacteral was the piper betle L. has antibacterial activity to Staphylococcus aureus.
