UJI AKTIVITAS ANTIBAKTERI FRAKSI N-HEKSAN DAUN SIRIH HIJAU (Piper betle L.) TERHADAP PERTUMBUHAN BAKTERI Staphylococcus aureus SECARA IN VITRO

- Background: One of the factors that may causes infection diseases is Staphylococcus aureus. The Staphylococcus aureus is a gram positive bacteria that can grow well at human body temperature as well as at food stored in room temperature. However, it can produce toxins at these temperatures. Objectives: The purposes of this research were to observe the antibacteria activity from Piper betle L. with methode n-hexane fraction an to determine the type of compound contained within n-hexane of Piper belte L. Method: To know the antibacterial activity, this research using disc diffusion method of n-hexane Piper betle L. The Piper betle L. fraction was obtained by using macerare method. In addition, the secondary determination metabolite of the plant was done by using the KLT method. The test concentration that used to test antibacterial activity was 250mg/ml, 500mg/ml, and 750mg/ml. The positive control used was Amoxiclav 30μg/disk, while for the negative control used was DMSO 1% + Tween 80 20% + sterile aquadest. The test liquid and test control were placed on the Mueller Hilton Agar media that had applied with bacterial suspension. Moreover, it was incubated at 37 o C for 24 hours. Result and Conclusion: The n-hexane fraction of Piper betle L. has antibacterial activity at concentration 250mg/ml has 8,88mm; 500mg/ml has 8,95mm and 750mg/ml 9,40mm μg/disk. On the positive control used Amoxiclav 30 μg/disk has 18, 65mm, while for negative control did not indicate the presence of the barrier zone. The compounds contained in the n-hexane fraction consist of alkaloid, terpenoids, flavonoids, polyphenols, and antrakuinon.
