- Background: Infection is one of the biggest factors that causes high number of illness and mortality. Staphylococcus aures is the bacterial which is frequently found in infection case. One of the plants that can be used to treat the infection is Piper betle L. Objective: This current study aimed to find out the bacterial activity by measuring inhibition zone diameter from ethanol fraction of Piper betle L. through the growth of Staphylococcus aureus and to find out the compound classification of the ethanol fraction. Method: Activity test of bacterial applied disc diffusion method. The ethanol fraction of Piper betle L. was obtained by conducting multilevel macerate and KLT test was conducted to indentify secondary metabolites compound. Result and Conclusion: Identification result of secondary metabolites showed that ethanol fraction of Piper betle L. contained alkaloids, terpenoids, polyphenols, and anthraquinone compound. In addition, the bacterial activity test result described that the average of inhibition zone diameter in concentration 250 mg/ml (12,5 mg/disc) has diameter 7,13 mm, concentration 500 mg/ml (25 mg/disc) has diameter 7,72 mm, and concentration 750 mg/ml (37,5 mg/disc) has diameter 8,48 mm, however positive control amoxicillin and clavulanic acid (30 μg/disc) was 24,92 mm. The conclusion of this reseacrh antibacterial was the average of inhibition zone diameter including the resistant category.
