- ABSTRACT Background: Staphylococcus aureus dan Streptococcus pyogenes are the kinds of gram-positive bacteria that become the main cause of pyoderma infection. The treatment for pyoderma is difficult because Staphylococcus aureus and Streptococcus pyogenes are resistant to some types of antibiotics. The basil leave (Ocimum basilicum) is known by having the antimicrobial effect because it has been proven that the basil leaves contains essential oil, tannins, saponins, alkaloids, and flavonoids. Objective: To prove that basil's extract has an antimicrobial effect on the growth of Staphylococcus aureus and Streptococcus pyogenes. Methods: This research used true experimental post-test only control group design with tube dilution method and the concentration of basil's extract of 100%, 50%, 25%, 12,5%, 6,25%, 3,125%, 1,56%, and 0%. The data was analyzed using the MANOVA and Post Hoc Games-Howell tests. Result and discussion: The Minimum Bactericidal Concentration (MBC) of Staphylococcus aureus is on 12,5% and 6,25% for Streptococcus pyogenes. The MANOVA test shows the significance of 0,000 sig <0,05 (p<0,05) which means that the basil’s extract has had an impact on the growth of both Staphylococcus aureus and Streptococcus pyogenes. The Post Hoc Games-Howell test shows that statistically, basil’s extract gives the same impact towards the growth of Staphylococcus aureus and Streptococcus pyogenes, while in the laboratory, the basil's extract has impacted Streptococcus pyogenes more. This matter is caused by the active ingredients of the basil itself such as essential oil, tannins, saponins, alkaloids, and flavonoids. Conclusion: The basil’s extract (Ocimum basilicum) has an antimicrobial effect on the growth of Staphylococcus aureus and Streptococcus pyogenes. Keywords: Basil’s extract (Ocimum basilicum), Staphylococcus aureus, Streptococcus pyogenes, Minimum Bactericidal Concentration (MBC).
