  • Background : Staphylococcus aureus is one of the bacteria that has been resistant to antibiotics. One of them is Methicillin-Resistant Staphylococcus aureus (MRSA). Ethyl acetate extract of Cerbera manghas L. fruit has the potential as an antibacterial that can inhibit the growth of Staphylococcus aureus. Objective : This study aims to determine the secondary metabolic compounds of ethyl acetate extract of Cerbera manghas L. fruit flesh and which compounds have antibacterial activity against Staphylococcus aureus using biaoutographic method. Method : This is an experimental research study to test the antibacterial activity of ethyl acetate extract of Cerbera manghas L. fruit against Staphylococcus aureus using bioautographic method by observing and measuring the diameter of inhibitory zone of each group of compounds in extracts that have been separated by TLC technique. Result : The results of this study showed that ethyl acetate extract of Cerbera manghas L. fruit have an alkaloid compounds at Rf = 0.47; steroid / terpenoids compounds at Rf = 0.64 and Rf = 0.82 with an inhibition area were about 6.04 ± 0.47 and 6.16 ± 0.68; flavonoid compounds at Rf = 0.52 with an inhibition area were about of 5.81 ± 0.12; polyphenols and tannins compounds at Rf = 0.36 with an inhibition area were about 5.39 ± 0.18. Conclusion : The inhibition area by the compounds of ethyl acetate extract of Cerbera manghas L. fruit had a significant difference when compared with the positive control cloramphenicol 30 μg / disk.
