- The decline in the quality of superior orchid seeds in Indonesia is due to the relatively small business scale, the lack of availability of selected superior seeds from cultivars, the price of superior seeds is relatively expensive, and the lack of market and capital information. One effort to increase the availability of superior seeds can be done by adding synthetic hormones into agar media used in In vitro culture. The purpose of this research was to determine the effect of auxin hormone addition on agar media on the growth of root length and the number of leaves on the moon orchid plants (Phalaenopsis amabilis). This research was conducted on May-June 2018 at DD ’Orchids Nursery, Kota Batu. The population in this study were some tissue culture bottles from the Genus Phalaenopsis that were 1-2 months old and were still in the germination stage in the first bottle. The sampling technique used in this research is simple random sampling. The data were analyzed using One Way Anova test by using SPSS 21 software. The results showed that the giving of auxin hormones with a concentration of 0.1 mg/l affected the growth of root length with an average value of 1.12, whereas the giving of auxin hormones with a concentration of 0.05 mg/l and 0.5 mg/l did not affect the root length. The giving of auxin hormones with concentrations of 0 mg/l, 0.05 mg/l, 0.1 mg/l and 0.5 mg/l did not significantly affect the growth of the number of leaves. The results of this research can be used as biology learning resource for twelfth high school students on the growth and development of living things in the form of handouts.
