- The number of apple trees in Batu city is 3,107,195 trees with a total production of 147,000 tons per year, but in 2004 there was a decline of 1 million trees which reduced local apple production by 46 thousand tons per year. Local farmers are still worried about the threat of weakening local apple production. Development of apple variations can be done by bringing new types of apple plants to Indonesia. The adaptation process to the tropical climate is not easy so that apple plants are not adaptable and die. One of the technological hopes for providing these plants is through Embryo culture techniques. Provision of plants in vitro through embryo culture has high success and can be grown directly through apple callus. In vitro plant propagation can be done through somatic embryogenesis either directly or callus induction first. The use of media with the composition of sucrose and amino acid treatment showed an interaction on the parameters of fresh callus weight, fresh weight of callus and the percentage of fresh weight gain of callus. In the sucrose composition 60 g / l + 150 mg / l CH + 100 mg / l Proline in solid media gave a tangible result to the parameters of callus fresh weight at age 8 MST of 0.73 g, the increase in fresh weight of callus was 0.26 g, and the percentage of fresh callus weight gain was 43.65%.
