APLIKASI GIBERELIN ACID (GA3) DAN INDOLE ACETIC ACID (IAA) TERHADAP PERTUMBUHAN DAN PERAKARAN TUNAS APEL MANALAGI SECARA KULTUR IN VITRO

- Propagation of Manalagi apple plant seeds that is done in a modern way is through in vitro culture. Problems that often arise in the process of apple shoot induction by in vitro method is that many explants tend to have short shoot sizes. In fact, to facilitate the subsequent culture process, explants in the form of shoots must be able to be distinguished between each stem segment and its leaves. Gibberelin plays a role in supporting cell ellongation, cambium activity and protein synthesis. While the addition of IAA exogenously will help endogenous auxin activity in inducing the root formation. This study aims to examine the effects of adding GA3 and IAA combinations to shoot growth and rooting of Manalagi apples by in vitro culture. The pre-study was conducted on January 9, 2018 until May 14, 2018. The study began on May 16, 2018 until June 27, 2018. Pre-research and research were conducted at the Mitra Anggrek Indonesia Culture Laboratory (MAI) St. Hasanudin Gg. 1 Numb. 9A Jeding Hamlet, Junrejo District, Batu City. The implementation of the study used a Completely Randomized Design (CRD) and repeated 10 times. The obtained data were analyzed by statistical analysis of variance (ANOVA) and a comparatively honest test of difference (BNJ) level of 5%. The addition of the combination of GA3 + IAA showed no significant effect on all variables. Both growth variables (number of shoots, length of shoots, number of branches, and number of leaves) and roots (number and length of roots). This is because the combination of the GA3 + IAA hormone tested has not shown the right concentration for shoot growth and rooting of Manalagi apple.
