- Background: Excitotoxicity that caused by excessive glutamate can disturb memory function by oxidative stress. Papaya leaves as neuroprotective contain flavonoid and fenolic that can inhibit free radicals caused by MSG. Objective: Determined the effect of papaya leaf extract to pyramidal cells of cortex cerebri and memory function of male white rat induced by MSG. Methods: Used true experimental method with post test only control group design approach. Male white rat of wistar strains grouped into 4 groups, K+ induced by MSG at a dose 0,8g/1ml/day, P1;P2;P3 induced by MSG and papaya leaf extract at a dose 30;60;120 (mg/kg/day) which used gastric sonde for 30 days. Memory function testing for all rats consist of adaptation and test phase then calculated based on score of false and correct when the rat entering arm of 8 Arm Radial Maze. Then calculated the number of picnotic pyramidal cells in 3 views. Result: Multivariate anova test was obtained significant influence between groups with value p=0,000, post hoc test obtained significant difference with value p=0,000 in each groups, pearson correlation test obtained strength value -0,974 and linear regression test obtained AR2=0,719 for picnotic pyramidal cells and AR2=0,765 for memory function score. Increased in number of picnotic pyramidal cells and decreased in memory function score because of flavonoid and fenolic that contained in papaya leaf extract could inhibit free radicals. Conclusion: Papaya leaf extract decrease the number of picnotic pyramidal cells on cortex cerebri and increase the memory function score of male white rat induced by MSG.
