  • In Indonesia Agave which is widely used for natural fiber is Agave sisalana. The demand for agave fiber from 2006 to 2009 tends to increase. Thus the development of agave plants in the country needs to be improved to meet industrial needs. Alternative propagation that can be implemented is through tissue culture with embryogenesis indirectly through the callus phase. This study uses Kinetin ZPT and 2,4-D which are expected to grow callus with a good profile and can grow fast. The study was conducted using factorial randomized block design with two treatments, namely the composition of the use of growth regulators in the form of kinetin and 2,4-D. Variable data can be analyzed and in the real honest appeal test (BNJ) level of 5%. Based on the results of the study, the results of the combination of kinetin composition treatment with the composition of 2.4 D showed a real interaction on the parameters when smoothing. In the composition of the media with Kinetin concentration of 3.5 ppm with 2.4-D (Dichlorophenoxyacetic Acid) 1 ppm or 3 ppm gave tangible results to the parameters when callus appeared ie 10.33 - 10.50 hsi. Kinetin concentration of 0.5 - 5 ppm gives the same effect on crumb and compact callus texture parameters with a mean of 52.78% - 63.89% and 41.67% - 47.22% in Agave sisalana callus induction. The concentration of 2,4 D (2,4-Dichlorophenoxyacetic Acid) 1-2 ppm gives the same effect on crumb and compact callus texture parameters with a mean of 54.17% - 60.42% and 39.58% - 45.83% in Agave sisalana callus induction.
