- In 2016 the data from (Statistics Indonesia 2017), showed that potato production was only 1.2 million tons / ha, but based on the Ministry of Agriculture (2016) statistical data, the availability of quality potato seeds in Indonesia was only 30% different from the need potatoes which are 300,000 tons / year (including imports). Based on these problems, this study was conducted with the aim to determine the optimization of media on multiplication of in vitro shoots of of potatoes (Solanum tuberosum L.) varieties of granola lembang and granola kembang. The study was conducted from February to May 2018 at the In Vitro (Biotechnology) Culture Laboratory of Muhammadiyah University of Malang with room temperature of 19-22 oC. This study used a Randomized Group Design (RAK) which was repeated 3 times. The first factor is the variety which consists of 2 levels, namely: V1 = Granola Lembang and V2 = Granola Kembang. The second factor is the media, which consists of 5 levels namely M1 = 0.005 mg / l NAA + 1 mg / l Kinetin, M2 = 0.5 mg / l IAA + 2 mg / l BAP, M3 = 0.2 mg / l GA3 + 2 mg / l Ca3 (PO4) 2 + 75 ml / l Coconut Water + 6 g / l Agar, M4 = 3 mg / l GA3 + 0.1 mg / l Kinetin, and M5 = 0.4 mg / l Tiamin + 2 mg / l Glycine + 2 mg / l Niacin + 1 mg / l IAA + 3 mg / l BAP + 0.5 mg / l GA3 + 0.03 mg / l NAA. Based on the results of the research optimization of media on multiplication of in vitro shoots of potatoes (Solanum tuberosum L.) varieties of granola lembang and granola kembang can be concluded that the varieties of Granola Lembang with media 0.2 mg / l GA3 + 2 mg / l Ca3 (PO4) 2 + 75 ml / l Coconut Water + 6 g / l To show the best results in shoot length, number of books, number of leaves, number of roots, and root length. In the potential for multiplication of shoots, media treatment 0.2 mg / l GA3 + 2 mg / l Ca3 (PO4) 2 + 75 ml / l Coconut water + 6 g / l In order to show the best results on varieties of granola lembang and granola kembang with multiplication value amounting to 620 and 299 planlets
