- Background: Infection is still a major problem in the health care. Natural resources has potential solution. Cerbera manghas L. seeds have antibacterial activity with active compounds such as alkaloids and steroids. Objective: The aim of this study was to determine the antibacterial activity ofextract n-hexane Cerbera manghas L.seeds against Staphylococcus aureus with bioautografi method and determine classes of compounds that provide antibacterial activity. Methods: Powdered Cerbera manghasL.seed extracted with 1000 ml n-hexane and macerated for 48 hours performed twice. Then extract the identification by TLC method and test the antibacterial activity using the contact bioautografi. Extract n-hexane Cerbera manghas L. seeds 500 mg hydrolyzed then spotted as many as 30 μl. There are 4 stains produced. The negative control used is sterile distilled water, whereas the positive control used chloramphenicol 30μg/disc. The bacterial suspension is applied on an agar medium has been solid. The test group, negative control and positive control placed on the media and incubated at 37o°C for 24 hours. The Results: Compounds identified by TLC method showed that the extract n-hexane Cerbera manghasL. seeds contain alkaloids and steroids. The results of the antibacterial activity testextract n-hexane Cerbera manghas L. seeds against Staphylococcus aureus as much as 30 μl give inhibition at Rf 1 = 6.49 mm, Rf 2 = 4.33 mm, Rf 3 = 6.49, Rf 4 = 8.03 mm and Rf 5 = 4.39 mm. It can be concluded that the extract n-hexane Cerbera manghas L. seeds possess antibacterial activity.
