UJI AKTIVITAS ANTIBAKTERI FRAKSI ETIL ASETAT UMBI Eleutherine palmifolia (L.) TERHADAP Escherichia coli DENGAN METODE DIFUSI CAKRAM

- Background: One of the infections that caused by Escherichia coli bacteria is diarrhea. Resistance with antibiotics oftenly happens, therefore need an alternative traditional treatment that is Eleutherine palmifolia L. Objective: the aim of this research is to find out the classes of secondary metabolites that found on ethyl acetate fraction at Eleutherine palmifolia L tubers and obtain data of inhibition zone ethyl acetate fraction Eleutherine palmifolia L to Escherichia coli growth. Method: to find out an antibacterial activity test using disc diffusion methods E. palmifolia tuber n-Hexane fraction with maceration level methods, then secondary metabolites identified with KLT methods and doing a test of bacterial activity with disc diffusion methods. The concentration of solvent test is 20 mg/ml, 40 mg/ml, and 60 mg/ml. Negative control is DMSO 1% and aquadest sterilization, while positive control is cefotaxime 30 μg/disc. Bacteria suspension swiped on solidify jelly as the media. Solvent test, negative controls and positive controls put on solidify media, then incubated on 37oC for 24 hours. Result and Conclusion: the result of secondary metabolites identification, ethyl acetate fraction of Eleutherine palmifolia L.contains alkaloid, flavonoid, terpenoid, polifenol, and antrakinon. The result of antibacterial activity testing of ethyl acetate fraction on Eleutherine palmifolia L tubers to Escherichia coli with 20 mg/ml(1,4 mg/disk) concentration has inhibition zone 20.5 mm, 40 mg/ml(2,8 mg/disk) concentration has inhibition zone 20.7 mm, and 60 mg/ml(4,2 mg/disk) concentration has inhibition zone 20.9 mm. Eventually, it can be concluded that ethyl acetate fraction of Eleutherine palmifolia L. tubers has an activity as antibacterial.
