  • Background : The one caused of diarrhea was an infection from Escherichia coli bacteria that was Gram negative bacteria. Part of Escherichia coli has been resistance to various antibiotics so it was necessary development of new bacteria which can against the infection. Objective : The aim of the study is to find out secondary metabolites which founded on the n-Hexane fraction of Eleutherine palmifolia and obtained the diameter inhibition zone of Eleutherine palmifolia n-Hexane fraction to Escherichia coli growths. Method : To find out an antibacterial activity testing that’s using disc diffusion methods. Eleutherine palmifolia tuber n-Hexane fraction with maceration level methods, then secondary metabolites identified with KLT methods and doing a bacterial activity testing with disc diffusion methods. The concentration used was 20 mg/ml, 40 mg/ml, and 60 mg/ml. negative control which used was DMSO 1% and aquadest sterilization, while positive control used was Cefotaxime 30μg/disc. Solution test and control test putt on the Mueller Hinton Agar media that has been spread bacteria suspension then incubated on 37 o C for 24 hours. Result and Conclusion : The result of the secondary metabolites identification with KLT was showing positive results on terpenoid in which it given purple spot and flavonoid given yellow intensive spot color. The result of antibacterial Eleutherine palmifolia n-Hexane fraction against Escherichia coli with 20 mg/ml, 40 mg/ml, 60 mg/ml concentrations didn’t give inhibition zone, it caused of diameter clear zone as biggest as disc diameter that was 6.0±0 mm. Then it will be concluded that was n-Hexane fraction of Eleutherine palmifolia has not biology activities as antibacterial.
