  • The availability of poor quality seed potatoes is a major factor in the production of potatoes in Indonesia. In 2008 the need of potato seeds was 96.277 tons, while the availability of domestic certified seeds was only 8.066 tons (8.3%). The solution of multiplication quickly of superior potato seeds is through stem cuttings. The purpose of this research was to test the differences in control and treatment of growth and production of potato seeds, to test the interaction of growing media and the number of segments and to obtain the appropriate growing media and number of segments for the growth and production of potato seed. The research was conducted at the Laboratory of Biotechnology University of Muhammadiyah Malang in March to August 2017, continued at Green House located in Pujon Kidul Village in September 2017 until January 2018. Research using Factorial Design (3 x 3) arranged in Random Block Design with three replications. The first factor is growing media with 3 levels, there are husk charcoal, cocopeat, humus. The second factor is the number of segments with 3 levels, there are three segments, four segments, five segments. Variant analysis was performed on observation result and continued by BNJ test with 5% level if there is real difference between treatment. Differences in the effect of control and treatment were analyzed using Orthogonal Contrast Test. The results showed that growing media and the number of segments affect the growth and production of potato seeds. There are differences in control and treatment of plant height, number of branches, diameter of the bearing, number of leaves, root length, wet weight of stalk, number of mini tubers per plant, tuber wet weight and dry weight of tuber. The influence of combination of humus growing media treatment with four segments to stem diameter. Charcoal husk growing media gives good result to total number of leaf corofil and root length, while humus growing media gives good result on wet weight of stalk, number of mini tubers per plant and dry weight of tuber. The four segment treatment gave good result to stem diameter while the five segment treatment gave good result on plant height, root length and wet weight of stem
