- Apple (Malus sylvestris Mill) is a perennial fruit plant originated from subtropical area of West Asia. Based on BPS Dirjen Horti (2016), there was an increase of apple productivity in 2015 by 68.556 tons or 65,26% compared in 2014, and number of imports in 2015 increased to 30,47% or 37,77 million US dollar. Low productivity of apple in Batu-Malang resulted by soil fertility degradation. Based on BPS Kota Batu (2010), there was a decrease in the number of apple production by 34,74%, that was from 1.291.352 quintals in 2009 to 842.799 quintals in 2010. One of solutions to boost local apple production is implementing in vitro culture. Plant preparation by in vitro culture through embryo culture is highly effective and able to directly cultivated as a shoot. In vitro shoot multiplication is purposed to multiply apple shoot that have been cultivated through subculture, and it is one of many ways to provide either germplasm or superior seed. Addition of different PGR compositions to culture media of apple shoot is effective and important. This study was aimed to examine the effect of PGR, TDZ, and IBA combination in Murashige Skoog (MS) medium to the apple shoot multiplication. It is expected that this study will be useful in development of plentiful apple varieties. This study was conducted using factorial Randomized Complete Block Design (RCBD) with two factors of treatment, they were apple varieties and TDZ and IBA combination, with three times repetition. The collected data was analyzed by variance analysis and Tukey multiple comparison test with 5% of α level. The result showed that combination of treatments in 8 WAP (week after planting) on Fuji variety with combination of 2,5 mg/l TDZ + 0,5 mg/l IBA in culture medium resulted higher number of shoots up to 22,20. Combination of Manalagi variety treatment with 2,5 mg/l TDZ + 0,5 mg/l IBA had higher enhancement of relative fresh weight up to 2,43 grams and higher relative explant height up to average 1,88 cm. Apple variety treatment of Fuji, Red Delicious dan Manalagi had the same ability in all observation variables. TDZ and IBA concentration treatment had real effect in number of shoots, enhancement of fresh weight, explant color percentage, explant height, leave number, sprouted explant, and time when shoot appear on combination of 2,5 mg/l TDZ + 0,5 mg/l IBA
