- Injection preparation was medicine by usage route critical enough if compared with oral preparation or other, because it directly connected with body tissue, then the injection preparation must be free from microorganism contamination. The aims of the study was defining the sterility of preparation injection difenhidramin HCl multiple dose with the preservatif chlorobutanol 0,5 % b/v toward the frequency retival in the repeated usage. In this study we used preparation injection difenhidramin HCl by frequency retival which was conducted 5 times based on the usage in the hospital, sample test was done in Laminar Air Flow Cabinet by direct inoculation method for 2 ml then incubated for 14 days and observed the results, first test we did the sterility test and fertility test of the medium, after that we did inactivation test of the preservatif with comparsion 1:4 for Thioglycollate and Kassamino and the last were sterility test of the preparation sample to be compared with the comparator control. From the result of this research by using preparation difenhidramin HCl with chlorobutanol 0,5% b/v with the effect of frequency retival for 5 days retipectively, it did not affter yhe sterility of preparation.
